{"id":17120,"date":"2018-03-09T09:05:03","date_gmt":"2018-03-09T09:05:03","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/identificacion-de-moleculas-de-interes-biomedico-a-partir-del-cribado-de-quimiotecas-combinatorias\/"},"modified":"2018-03-09T09:05:03","modified_gmt":"2018-03-09T09:05:03","slug":"identificacion-de-moleculas-de-interes-biomedico-a-partir-del-cribado-de-quimiotecas-combinatorias","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/identificacion-de-moleculas-de-interes-biomedico-a-partir-del-cribado-de-quimiotecas-combinatorias\/","title":{"rendered":"Identificaci\u00f3n de mol\u00e9culas de inter\u00e9s biom\u00e9dico a partir del cribado de quimiotecas combinatorias"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Teresa Carbonell Castell\u00f3 <\/strong><\/h2>\n<p>Utilizando la qu\u00edmica combinatoria, se han sintetizado dos quimiotecas de mol\u00e9culas discretas, 2,5-piperazindionas (dkps) (105 mol\u00e9culas) y dos quimiotecas combinatorias en formato de rastreo posicional, una de pseudop\u00e9ptidos (10,648 mol\u00e9culas) y otras de haxap\u00e9ptidos de d-amino\u00e1cidos con el extremo terminal acetilado (47,045,881 mol\u00e9culas).  esta diversidad qu\u00edmica es la que se ha utilizado en la identificaci\u00f3n de moduladores de tres actividades enzim\u00e1ticas, relacionadas con distintas patolog\u00edas, y en la identificaci\u00f3n de moduladores del crecimiento bacteriano. para ello, se desarrollaron los ensayos biol\u00f3gicos adecuados para cada una de las dianas, y a continuaci\u00f3n se exponen brevemente los resultados:  &#8211; en el caso de las acetilcolinesterasa (ache), enzima importante por su implicaci\u00f3n en la enfermedad de alzeimer, se han identificado distintas dkps con una potencia y selectividad en la inhibici\u00f3n de la misma elevada. concretamente, destacar el compuesto dkp-ii 17, que pude considerarse para el estudio de optimizaci\u00f3n de sus propiedades.  &#8211; para la proteasa del virus de la inmunodeficiencia adquirida humana (hiv-1-pr), cuya funci\u00f3n es esencial en el ciclo vital de este virus que ocasiona el sida, se han identificado distintos hexap\u00e9ptidosque la inhiben (constantes de inhibici\u00f3n en el intervalo micromolar). De entre ellos, destacar los hexap\u00e9ptidos hxah10, hxah13 y hxah14, que presentan constantes de inhibici\u00f3n entre 1-2 micromolar e interaccionan de forma espont\u00e1nea con la proteasa, seg\u00fan indican estudios de calorimetr\u00eda de titulaci\u00f3n isot\u00e9rmica.  &#8211; tambi\u00e9n se han identificado inhibidores potentes de la plasmepsina ii(plmii), enzima esencial para el ciclo vital de par\u00e1sitos del g\u00e9nero plasmodium, agentes causantes de la malara. Concretamente, todos los pseudop\u00e9ptidos y hexap\u00e9ptidos identificados, presentan una inhibici\u00f3n en el intervalo micromolar, destacando como el inhibidor m\u00e1s potente y
