Este trip\u00e9ptido es esencial para la protecci\u00f3n frente a los radicales libres y los ataques electrofilicos y, por ello, regula la sensibilidad celular a las radiaciones y los f\u00e1rmacos citot\u00f3xicos. La depleci\u00f3n de gsh inhibe el crecimiento tumoral in vitro e in vivo. Nuestro grupo de investigaci\u00f3n demostr\u00f3 que la depleci\u00f3n selectiva de gsh tumoral in vivo y la radioterapia son capaces de eliminar un c\u00e1ncer de crecimiento local y agresivo como es el tumor asc\u00edtico de ehrlich (estrela y cols., 1995), pero el mismo protocolo experimental aplicado en un modelo experimental de met\u00e1stasis como el melanoma murino b16 se mostr\u00f3 ineficaz. Por ello nos propusimos estudiar en profundidad el metabolismo del gsh en las c\u00e9lulas metast\u00e1ticas in vitro, su posible relaci\u00f3n con la capaciad de met\u00e1stasis hep\u00e1ticas in vivo, as\u00ed como el papel del gsh en la protecci\u00f3n contra el ataque oxidativo y nitrosativo del endotelio sinosoidal hep\u00e1tico. Nuestros resultados demuestran que: a) las celulas de mb16 con un elevado contenido en gsh poseen una mayor capacidad metast\u00e1tica in vivo. por lo tanto, el gsh constituye un par\u00e1metro celular importante para la progresi\u00f3n metast\u00e1tica. B) el no y el h2o2 son factores cr\u00edticos en la actividad antitumoral del endotelio sinusoidal hep\u00e1tico frente a las c\u00e9lulas potencialmente metast\u00e1sicas. Esta citotoxicidad tumoral es dependiente de la producci\u00f3n de no, el cual es particularmente t\u00f3xico en presencia de h2o2. C) las c\u00e9lulas del mb16 con un elevado contenido en gsh son mucho m\u00e1s resistentes a la citotoxicidad inducida por el no y el h2o2. Las c\u00e9lulas tumorales sometidas a esta situaci\u00f3n de estr\u00e9s dependen fundamentalmente del gsh y la gpx para detoxificar el h2o2.
