La amplia variedad de complejos y condiciones experimentales en los que se pueden efectuar los an\u00e1lisis la convierten en una t\u00e9cnica estructural atractiva. Sin embargo, esta metodolog\u00eda se enfrenta a graves dificultades en la calidad de las im\u00e1genes obtenidas. las presente tesis aborda esta cuesti\u00f3n desde una perspectiva de procesamiento digital de im\u00e1genes. El problema resuelto es la identificaci\u00f3n de una estructura tridimensional compatible con el conjunto de proyecciones medido con el microscopio, empleando para ello una t\u00e9cnica iterativa de tomograf\u00eda tridimensional que opera en el espacio real (algebraic reconstruction technique, art). tres han sido los pilares de la presente tesis: por una parte, la optimizaci\u00f3n sistem\u00e1tica de los par\u00e1metros implicados en la reconstrucci\u00f3n; por otra, la correcci\u00f3n de la funci\u00f3n de transferencia del microscopio (llamada en el campo ctf, contrast transfer function); por \u00faltimo, la inclusi\u00f3n de informaci\u00f3n a priori en el propio algortimo de reconstrucci\u00f3n. La combinaci\u00f3n de estos tres factores ha proporcionado importantes mejoras, superiores a 10-15 angstroms de resoluci\u00f3n, tal y como lo demuestran los experimentos llevados a cabo tanto con vol\u00famenes artificiales, phantoms, como con prote\u00ednas reales. Adicionalmente se ha extendido el m\u00e9todo de reconstrucci\u00f3n en el espacio real para el tratamiento de im\u00e1genes de cristales biol\u00f3gicos.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
