Las orexinas activan dos receptores de membrana acoplados a prote\u00ednas g, denominados receptor 1 de orexina (ox1r) y receptor 2 de orexina (ox2r).  las implicaciones fisiol\u00f3gicas de este sistema son m\u00faltiples y variadas, y sugieren que las orexinas pueden actuar como nexo de uni\u00f3n entre procesos aparentemente tan dispares, como la ingesta y el ciclo sue\u00f1o-vigilia.  el objetivo de este trabajo ha sido estudiar las acciones neuroendocrinas de la orexina-a, y los mecanismos de regulaci\u00f3n de la expresi\u00f3n de prepro-orexina, con el fin de ayudar a esclarecer la fisiolog\u00eda de este sistema ligando-receptor.  los resultados obtenidos nos permiten concluir que:  1,- la orexina-a estimula la ingesta. Esta acci\u00f3n parece estar mediada por la regulaci\u00f3n de la expresi\u00f3n de npy en el hipot\u00e1lamo, pero no de otros p\u00e9ptidos como agrp, mch, cart o crh.  2,- la orexina-a regula los niveles de mrna de ghrh en el n\u00facleo paraventicular del hipot\u00e1lamo, pero no los modifica en el n\u00facleo arcuato. La orexina-a regula la expresi\u00f3n de somatostatina en el n\u00facleo periventricular del hipot\u00e1lamo, en ausencia de hormona de crecimiento.  3,- el ayuno regula la expresi\u00f3n de prepro-orexina y del receptor 1 de orexinas en el hipot\u00e1lamo, efecto revertido por la leptina. El efecto del ayuno es tejido-espec\u00edfico, dado que la expresi\u00f3n de los receptores de orexina en la gl\u00e1ndula adrenal no se ve afectada por esta situaci\u00f3n.  4,- la expresi\u00f3n de prepro-orexina en el hipot\u00e1lamo lateral no est\u00e1 regulada ni por hormonas tiroideas, ni por glucocorticoides ni por hormona de crecimiento, ni por ghrh.  5,- el incremento de la ingesta neonatal induce un fenotipo obeso hasta la etapa adulta.
