Luisa <\/strong><\/h2>\n<p>Cot\/tpl-2 quinasa es una map quinasa quinasa quinasa implicada en activaci\u00f3n celular y proliferaci\u00f3n. La deleci\u00f3n del extremo carboxi-terminal de cot\/tpl-2 desenmascara el potencial oncog\u00e9nico de la prote\u00edna, sin embargo, por el momento se desconoce el mecanismo por cual se produce dicha transformaci\u00f3n.  en esta tesis doctoral, demostramos que la deleci\u00f3n del extremo carboxi-terminal de cot wt incrementa la actividad cot quinasa total mediante dos mecanismos diferentes. Cot truncada tiene una actividad quinasa espec\u00edfica 3,9 veces mayor que cot wt. Adem\u00e1s, la prote\u00edna cot truncada, con una vida media de 95 min., Tiene una expresi\u00f3n en estado estacionario 2,6 veces mayor que cot wt, que muestra una vida media de 35 min. La combinaci\u00f3n de estos dos sucesos, desencadena una actividad cot quinasa espec\u00edfica total 10 veces mayor en las c\u00e9lulas transfectadas con cot quinasa truncada  comparadas con las transfectadas con cot wt.  en este trabajo adem\u00e1s demostramos que tanto cot wt como cot truncada, son degradadas v\u00edas proteasoma en c\u00e9lulas intactas de un modo independiente de la ubiquitina. El proteasoma 20s degradada adem\u00e1s, de forma m\u00e1s eficiente, a cot wt que a cot truncada traducidas y transcritas \u00abin vitro\u00bb, y lo que es m\u00e1s, el proteasoma tambi\u00e9n se encarga de la degradaci\u00f3n de las formas inactivas de ambas quinasas. Por otro lado, la inserci\u00f3n del extremo carboxi-terminal de cot wt en una prote\u00edna amarilla fluorescente (eyfp) confiere inestabilidad a la prote\u00edna y provoca la degradaci\u00f3n de la prote\u00edna de fusi\u00f3n peyfp-c-cot 390-466 por el proteasoma. La prote\u00edna gst-c-cot 390-466 es un sustrato para el proteasoma 20s \u00abin vitro\u00bb.
