Api se sintetiza en el citoplasma y es transportada a la vacuola en el interior de ves\u00edculas llamadas ves\u00edculas cvt o autofagosomas, seg\u00fan las condiciones metab\u00f3licas en que se encuentre la c\u00e9lula. El extremo amino terminal de la prote\u00edna api es determinante para su localizaci\u00f3n en estas ves\u00edculas. Para encontrar prote\u00ednas implicadas en este sistema de transporte, decidimos buscar aqu\u00e9llas que interaccionaran in vitro con el extremo amino terminal de api, o in vivo, utilizando el sistema de doble h\u00edbrido.  en el estudio in vitro se detect\u00f3 la interacci\u00f3n de las chaperonas citos\u00f3licas de la familia de hsp70, ssa1p y ssa2p, con la regi\u00f3n amino terminal. Mediante el uso de un mutante portador de un alelo termosensible del gen ssa1 y carente del resto de3 los genes ssa, se pudo demostrar que la prote\u00edna ssa1p es requerida in vivo para el transporte de api a la vacuola en un paso previo a la formaci\u00f3n de las ves\u00edculas transportadoras.  utilizando el sistema de doble h\u00edbrido, confirmamos y caracterizamos la interacci\u00f3n entre api y una prote\u00edna, codificada por el gen yol082w, entonces de funci\u00f3n desconocida. El estudio del fenotipo de un mutante en dicho gen permiti\u00f3 demostrar la implicaci\u00f3n de esta prote\u00edna, que se denomin\u00f3 cvt19p, en la ruta de api a la vacuola.
