Para ello se ha utilizado el sistema del doble h\u00edbrido en levaduras basado en el factor de transcripci\u00f3n gal4. Se dispon\u00edan de dos genotecas de cdna de plantas (arabidopsis thaliana y nicotina bethamiana) fusionadas al dominio de activaci\u00f3n de gal4.  se clon\u00f3 el gen c1 de tylcsv en tres plasmidos cebo distintos, uno de baja expresi\u00f3n (pbd-gal4) y dos de alta expresi\u00f3n (pas2 y pgbkt7) pero con caracter\u00edsticas funcionales diferentes. Los resultados obtenidos han sido diferentes dependiendo del pl\u00e1smido utilizado para expresar la prote\u00edna cebo. La estirpe de s.Cerevisiae utilizada para realizar las b\u00fasquedas (pj696), posee tres genes chivatos para la interacci\u00f3n entre las dos prote\u00ednas de fusi\u00f3n: his3, ade2 y lacz. S\u00f3lo consideramos interacciones positivas, aquellas que eran capaces de activar los tres genes chivatos de la levadura (positivos primarios). Una vez obtenidos lo positivos primarios comprob\u00e1bamos:  a,- la existencia de la interacci\u00f3n retransformando levaduras con el clon de la genoteca y el pl\u00e1smido cebo.  b,- su especificidad, comprobando que el clon no activa por si s\u00f3lo.  c,- la naturaleza del clon, sucuenci\u00e1ndolo.  en las b\u00fasquedas detectamos tres tipos de falsos positivos:  a,- clones que no se confirmaron cuando se retransformaron levaduras.  b,- clones que activaban por si solos los genes chivatos.  c,- clones que recuperaban el fenotipo de interacci\u00f3n, no activaban por si solos, pero cuya secuencia correspond\u00eda a un polia.  la mayor\u00eda de los falsos positivos obtenidos fueron de tipo (a) en las b\u00fasquedas con el pl\u00e1mido pas2 y de tipo (b) en las b\u00fasquedas realizadas con pgbkt7.  el gen ubc9 de n.Benthamiana (nbubc9), aislado en este trabajo mediante el sistema del doble h\u00edbrido por su i<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
