Al ser el paso que catalizan limitante en ambas v\u00edas, representa un excelente punto de control estando sometidas a una fuerte regulaci\u00f3n gen\u00e9tica, alost\u00e9rica y por fosforilaci\u00f3n. En escherichia coli una sola cps sintetiza el carbamil fosfato utilizado en ambas rutas, siendo este enzima bacteriano inhibido por retroalimentaci\u00f3n por ump y activado por ornitina, imp y amonio. En mam\u00edferos la cps encargada de la bios\u00edntesis de primidinas (cpsii) forma parte de un polip\u00e9ptido multienzim\u00e1tico de 243 kda llamado prote\u00edna cas, que alberga las tres primeras actividades enzim\u00e1ticas de la ruta (cpsii-aspartato   transcarbamilasa y dihidroorotasa). la cpsii de cad se activa por fosforribosilpirofosfato y se inhibe por utp estando, adem\u00e1s, la regulaci\u00f3n alost\u00e9rica controlada por fosforilaci\u00f3n por la prote\u00edna quinasa dependiente de amp c\u00edclico (pka) y por la prote\u00edna quinasa activada por mit\u00f3genos (mapk).  mientras se conoce la estructura tridimensional del enzima bacteriano, no se dispone de informaci\u00f3n estructural sobre la cpsii de la prote\u00edna cad. Sobre la base de resultados de nuestro laboratorio y de otros grupos proponemos para el sitio de nucle\u00f3tido efector una similar organizaci\u00f3n estructural en todas las cpss involucradas en la bios\u00edntesis de pirimidinas. aqu\u00ed ponemos a prueba esta propuesta mediante mutag\u00e9nesis dirigida al sitio de fosforilaci\u00f3n por pka que controla la uni\u00f3n de nucle\u00f3tido efector en cpsii. El sitio de fosforilaci\u00f3n para pka en la prote\u00edna cad no est\u00e1 presente en la cps bacteriana, por lo que lo hemos introducido en la enzima de e.Coli, debido a la imposibilidad de disponer de prote\u00edna cad recombinante funcional con la que realizar el estudio de mutag\u00e9nesis.
