El objetivo de este trabajo es en primer lugar, establecer si igf-i y vip tienen acci\u00f3n mitotr\u00f3fica sobre un tipo celular concreto, las c\u00e9lulas lactotropas hipofisarias. En segundo lugar definir los mecanismos de se\u00f1alizaci\u00f3n de igf-i y vip en su acci\u00f3n proliferativa y los mecanismos de la interacci\u00f3n en las rutas de se\u00f1alizaci\u00f3n de ambos factores. Hemos establecido la acci\u00f3n de igf-i y vip sobre la proliferaci\u00f3n de las c\u00e9lulas lactotropas. En estudios previos hemos evidenciado que tanto igf-i como vip son factores liberadores de prolactina y adem\u00e1s que la cci\u00f3n de igf-i est\u00e1 mediada por el vip hipofisario cuya expresi\u00f3n induce. Tambi\u00e9n hemos observado que ambos factores tienen una acci\u00f3n mitotr\u00f3fica sobre las c\u00e9lulas lactotropas  y que igf-i necesita la presencia de vip para ejercer totalmente su acci\u00f3n proliferativa. Igf-i ejerce sus acciones proliferativas a trav\u00e9s de la ruta de se\u00f1alizaci\u00f3n ras\/mapk\/erk mientras que vip se\u00f1aliza a trav\u00e9s de la v\u00eda ampc\/pka. Nuestros resultados indican la dependencia de igf-i en la activaci\u00f3n de la v\u00eda de se\u00f1alizaci\u00f3n ampc\/pka para inducir su acci\u00f3n mitotr\u00f3fica. Adem\u00e1s, vip es capaz de inducir la activaci\u00f3n de la v\u00eda mapk, necesaria para promover la expresi\u00f3n g\u00e9nica inducida por vip y la proliferaci\u00f3n celular. Igf-i y vip inducen la expresi\u00f3n de ciertos genes a trav\u00e9s de la activaci\u00f3n de factores transcripcionales como creb el cual se une al elemento cre que est\u00e1 presente en el promotor de numerosos genes de expresi\u00f3n temprana y de expresi\u00f3n tard\u00eda implicados en la proliferaci\u00f3n celular. Igf-i y vip requieren la activaci\u00f3n de las v\u00edas de se\u00f1alizaci\u00f3n mapk y ampc\/pka para ejercer sus acciones mitog\u00e9nicas sobre las c\u00e9lulas lactotropas.
