{"id":18633,"date":"2018-03-09T09:07:15","date_gmt":"2018-03-09T09:07:15","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/clonacion-y-expresion-de-los-genes-clt-de-myxococcus-xanthus-con-capacidad-coagulante-de-leche\/"},"modified":"2018-03-09T09:07:15","modified_gmt":"2018-03-09T09:07:15","slug":"clonacion-y-expresion-de-los-genes-clt-de-myxococcus-xanthus-con-capacidad-coagulante-de-leche","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-de-la-vida\/clonacion-y-expresion-de-los-genes-clt-de-myxococcus-xanthus-con-capacidad-coagulante-de-leche\/","title":{"rendered":"Clonaci\u00f3n y expresi\u00f3n de los genes clt de myxococcus xanthus con capacidad coagulante de leche"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Margarita Poza Dom\u00ednguez <\/strong><\/h2>\n<p>Myxcoccus xanthus es capaz de secretar gran cantidad de mol\u00e9culas de inter\u00e9s industrial tales como antibi\u00f3ticos, carotenoides y enzimas hidrol\u00edticos. anteriores estudios indican que podr\u00edan existir de proteasas producidas por m.Xanthus de distinta naturaleza bioqu\u00edmica.  bas\u00e1ndonos en trabajos que exponen la idea de que la mixobacteria podr\u00eda ser una excelente fuente de proteasas de inter\u00e9s biotecnol\u00f3gico, en concreto, en la industria quesera, capaces de coagular la leche, se plante\u00f3, en primer lugar la caracterizaci\u00f3n de las proteasas producidas por 4 cepas diferentes de m.Xanthus as\u00ed como su posterior producci\u00f3n semi-industrial empleando una planta piloto de fermentaci\u00f3n.  as\u00ed, se describi\u00f3 que los sobrenadantes de los cultivos de las cepas de m.Xanthus 422, dk1622, dx10407 y dzf1, muestran actividad coagulante sobre leche entera y desnatada, siendo la cepa de m.Xanthus 422 la que mostr\u00f3 los valores de actividad m\u00e1s altos. La actividad m\u00e1xima se observa en todas las cepas al final de la fase exponencial. Las condiciones \u00f3ptimas para el crecimiento de las cuatro cepas y la producci\u00f3n de la proteasa coagulante fueron de 33\u00c2\u00bac y 200 rpm, en el medio ctt. Adem\u00e1s de la actividad coagulante, las cepas de m.Xanthus 422, dk1622, dzf1 y dk10407 producen otras proteasas degradativas capaces de originar halos en placas de agarosa con case\u00edna y de deshacer el co\u00e1gulo l\u00e1cteo. Esta actividad se detecta directamente en el sobrenadante de la cepa dzf1 o en los concentrados de los sobrenadantes del resto de las cepas. El factor de concentraci\u00f3n m\u00e1ximo para la obtenci\u00f3n de los concentrados de cuajo fue de 20 veces. Por encima de este valor, la actividad no aumenta y se favorece la acci\u00f3n de las proteasas degradativas que deshacen la cuajada. La cepa de m.Xanthus 422 produce una proteasa coagulante de aproximadamente 40 kda, con un ph \u00f3ptimo de 6, una temperatura \u00f3ptima de actuaci\u00f3n de 37\u00c2\u00bac y un punto
