A continuaci\u00f3n sub\u00f3ptimas de arginina se generan ani\u00f3n super\u00f3xido y \u00f3xido n\u00edtrico por la nos. Ambos radicales libres reaccionan r\u00e1pidamente originando ani\u00f3n peroxinitrito (onoo), un compuesto inestable, t\u00f3xico para neuronas pero no para astrocitos. Nosotros pensamos que el onoo puede tener un papel fisiol\u00f3gico doble actuando como se\u00f1al moduladora que facilita la liberaci\u00f3n de arginina por los astrocitos y adem\u00e1s previniendo la formaci\u00f3n de onoo en neuronas tras la activaci\u00f3n de la nnos.  por ello, estudiamos la regulaci\u00f3n del transporte de l-(3h)arginina tras un aporte ex\u00f3geno o end\u00f3geno de peroxinitrito en c\u00e9lulas gliales.  la liberaci\u00f3n de l-arginina mediada por peroxinitrito aument\u00f3 dos veces en c6 de forma sodio-independiente. Este incremento se previno por 5mm de l-arginina en ausencia de sodio y por 5 mm de l-alanina o l-leucina en presencia de sodio. El aumento en la captaci\u00f3n de l-arginina mediada por peroxinitrito fue transestimulada por 10 mm de l-arginina e inhibida de manera concentraci\u00f3n dependiente por el inhibidor del sistema y+, la n-etilmaleimida, en c6.  el peroxinitrito formado end\u00f3genamente en astrocitos tratados con lps, aument\u00f3 el transporte de l-arginina sin alterar los niveles del rna del cat-1. An\u00e1lisis de western blot de astrocitos y c6 tratadas con onoo revelaron una banda 3-nitrotirosinada anti-cat1-inmunopositiva, surgiriendo fuertemente la nitraci\u00f3n del transportador cat1 como causa de su activaci\u00f3n. Adem\u00e1s, el onoo no alter\u00f3 el transporte de l-arginina en fibroblastos carentes (homozig\u00f3ticos) del gen que codifica para cat1. Por \u00faltimo, l-arginina liberada por astrocitos tratados con onoo-, fu\u00e9 eficazmente captada por las neuronas en sistemas de cocultivos.
