Algunos tipos celulares son capaces de escapar a dicho control durante su programa de diferenciaci\u00f3n terminal. Este es el caso del megacariocito, la c\u00e9lula precursora de las plaquetas sangu\u00edneas.  en las \u00faltimas etapas de su maduraci\u00f3n, el megacariocito adquiere el fenotipo poliploide por establecimiento de ciclos de endorreplicaci\u00f3n, o endomitosis, consistentes en repetidas fases s sin divisi\u00f3n celular concomitante. duante la endorreplicaci\u00f3n megacarioc\u00edtica, los complejos ciclinae\/cdk2, responsables de la transici\u00f3n g1\/s, se mantienen activos. Nos preguntamos si los inhibidores de estos complejos, p21cip1 y p27kip1 estaban implicados en la regulaci\u00f3n de la endorreplicaci\u00f3n. El sistema experimental que utilizamos ha consistido en comparar dos l\u00edneas celulares megacariobl\u00e1sticas, hel y k562, ya que ambas diferencian hacia megacariocito en respuesta al tratamiento con \u00e9steres de frobol (tpa), pero s\u00f3lo en las hel se produce endorreplicaci\u00f3n. la expresi\u00f3n de p21cip1 se indujo a tiempos tempranos de la diferenciaci\u00f3n de ambas c\u00e9lulas, decayendo posteriormente de manera independiente a la capacidad de establecer o no endorreplicaci\u00f3n. Consecutivamente, los niveles de expresi\u00f3n de p27kip1 aumentaron en ambas l\u00edneas celulares. No obstante, los niveles de p27kip1 y del correspondiente complejo p27kip1\/ciclinae fueron en todo momento superiores en las c\u00e9lulas k562 que no endorreplican. estos datos sugieren que el equilibrio entre inhibidor y ciclina, m\u00e1s que la ausencia\/presencia de p27kip1, puede ser determinante para el establecimiento de la endorreplicaci\u00f3n. De hecho, cuando los niveles relativos de estos factores se alteraron por expresi\u00f3n constitutiva de ciclina e en las c\u00e9lulas k562, o expresi\u00f3n inducida de p27kip1 en las
