A pesar de todo, los mecanismos de transducci\u00f3n que controlan la liberaci\u00f3n i la s\u00edntesis de histamina son poco conocidos. En este sentido, se ha descrito que los autoreceptores h3 tienen una funci\u00f3n principal en la regulaci\u00f3n de los niveles de histamine cerebral pero, hasta el d\u00eda de hoy, los mecanismos de transducci\u00f3n de se\u00f1al acoplados a estos autoreceptores son poco conocidos.  la principal finalidad de este trabajo ha sido la descripci\u00f3n de las vias de transducci\u00f3n de se\u00f1al involucradas en la modulaci\u00f3n de la liberaci\u00f3n i la s\u00edntesis de histamine mediante los autoreceptores h3.  hemos desarrollado un m\u00e9todo radioisotopico para purificar por cromatograd\u00eda l\u00edquida de elevada resoluci\u00f3n (hplc) [3h]-histamina sintetizada a partir de histidina radioactiva lo cual permite una cuantificaci\u00f3n de la [3h]-histamina m\u00e1s senzilla, sensible y exacta que la obtenida mediante otros m\u00e9todos descritos previamente.  hemos demostrado que los autoreceptores h3 modulan la s\u00edntesis de [3h]-histamina mediante la via de la adenilato ciclasa-proteina quinasa a.  a pesar de ello, basandonos en nuestros resultados no podemos concluir si esta \u00faltima proteina quinasa activa a la histidina descarboxilasa de forma directa mediante la fosforilaci\u00f3n del enzima o de forma indirecta mediante la activaci\u00f3n de otras proteinas quinasa intermediarias.  por otro lado, hemos observado que los autoreceptores h3 no pueden modular la liberaci\u00f3n de [3h]-histamina mediante la via de transudcci\u00f3 que hemos citado previamente.  tambi\u00e9n hemos descrito que los incrementos de la liberaci\u00f3n i la s\u00edntesis de [3h]-histamina inducidos por despolarizaci\u00f3n con potasio est\u00e1n mediados por el incremento de la entrada de calcio extracelular a trav\u00e9s de canales de calcio activados por voltage de tipo n.
