La prote\u00edna codificada por ydl097c, denominada rpn6, es una subunidad reguladora del protesoma 26s, proteasa celular responsable de la degradaci\u00f3n citoplasm\u00e1tica. se generaron cepas de levadura en las que la copia gen\u00f3mica de rpn6 est\u00e1 eliminada (rpn6-a1), de forma que las c\u00e9lulas mutantes expresan condicionalmente la prote\u00edna en un medio de cultivo adecuado. En ausencia de rpn6p se produce una acumulaci\u00f3n progresiva de prote\u00ednas multiubiquitinadas, y la prote\u00edna hom\u00f3loga s9 del proteasoma humano complementa la mutaci\u00f3n rpn6-a1. Tras incubaciones prolongadas en el medio restrictivo, las c\u00e9lulas mutantes en rpn6 cesan su crecimiento, deteni\u00e9ndose en metafase.  las c\u00e9lulas muestra un fenotipo terminal uniforme, caracter\u00edstico de una parada en metafase: c\u00e9lulas dobles que no han sufrido citocinesis, contenido 2c de adn, n\u00facleo no dividido con un huso mit\u00f3tico corto e intranuclear, y elevados niveles de actividad quinasa asociada a cdc28. Asimismo, los sustratos de la enzima ligasa apc, pds1 y clb2, se acumulan en las c\u00e9lulas de cepa mutante condicional en condiciones restrictivas. La funci\u00f3n de rpn6 es esencial en mitosis, en concreto para que tenga lugar la anafase.  por otra parte, se purificaron proteasomas a partir de las c\u00e9lulas de la cepa rpn6-a1 mediante cromatograf\u00eda de afinidad. Los proteasomas obtenidos muestran una actividad peptidasa inferior a la de proteasomas silvestres, mientras que su actividad atpasa es id\u00e9ntica. Adem\u00e1s, los proetasomas mutantes presentan una movilidad electrofor\u00e9tica mayor en geles no desnaturalizantes que los proteasomas silvestres, siendo su coeficiente de sedimentaci\u00f3n en gradientes de densidad menor.  asimismo, los proteasomas obtenidos a partir de las c\u00e9lulas de la cepa rpn6-a1 han perdido las subunidades que componen la tapa del complejo
