En el primer trabajo de la tesis se estudian tres sistemas de producci\u00f3n recombinante: un sistema eucariota (pichia pastoris) y dos sistemas procariotas (escherichia coli). Se optimizan varios par\u00e1metros para la mejora de su nivel de producci\u00f3n y se buscan sistemas que permitan simplificar su proceso de purificaci\u00f3n. E.Coli con una producci\u00f3n final de prote\u00edna que es 10 veces superior al sistema de partida.  en el segundo trabajo se analiza el papel de las regiones precursora del pci en el plegamiento de la forma madura. Se analiza el proceso de replegamiento oxidativo in vitro de las distintas pro-formas del pci para caracterizar su influencia sobre la velocidad o eficiencia de plegamiento de la forma madura. Tambi\u00e9n se estudia la influencia de la pro-regi\u00f3n n-terminal sobre el proceso de plegamiento in vivo en e.Coli del pci maduro y de una serie de mutantes que presentan un mal plegamiento o un bajo nivel de expresi\u00f3n recombinante. Finalmente se lleva a cabo una caracterizaci\u00f3n estructural de la forma correspondiente al pci con la pro-regi\u00f3n n-terminal a trav\u00e9s de estudios de intercambio prot\u00f3n\/deuterio, exoprote\u00f3lisi limitada, dicro\u00edsmo circular y resonancia magn\u00e9tica nuclear.  en el \u00faltimo trabajo se analiza el proceso de plegamiento proteico de la hirudina y del lci mediante el estudio de las reacciones de intercambio prot\u00f3n\/deuterio seguidas por espectrometr\u00eda de masas maldi-tof. Se caracteriza la adquisici\u00f3n de estabilidad conformacional durante el proceso de plegamiento de las dos prote\u00ednas.
