En esta tesis hemos demostrado la existencia de un \u00abcross-talk\u00bb transcripcional entre las hormonas tiroideas y el oncog\u00e9n ras. En c\u00e9lulas de neuroblastoma n2a-beta que expresan de forma estable la isoforma beta del receptor de t3 (tr), t3 bloquea la proliferaci\u00f3n inducida por ras oncog\u00e9nico. Una de las dianas principales de los efectos proliferativos y transformantes de ras es la ciclina d1. T3 reduce fuertemente la expresi\u00f3n de esta ciclina inducida por ras, al mismo tiempo que incrementa los niveles de los inhibidores de complejos ciclina-cdk p21 y p27. Ras activa la transcripci\u00f3n de la ciclina d1 a trav\u00e9s de la activaci\u00f3n de los factores creb y atf-2 que se unen a un elemento de respuesta a ampc (cre) localizado en el promtor proximal. esta activaci\u00f3n ocurre v\u00eda raf\/mek\/erks. T3 bloquea la estimulaci\u00f3n del promotor de la ciclina d1 inducida por ras, raf o erk, pero no por pi3k o rsk2, una diana de las erks. La represi\u00f3n por t3 no est\u00e1 mediada la uni\u00f3n del receptor a un elemento de respuesta negativo (tre) sino a trav\u00e9s del antagonismo transcripcional con el sitio cre por represi\u00f3n de la actividad transcripcional de creb y atf-2.  t3 tambi\u00e9n bloque la actividad quinasa de rsk2 inducida por ras, sugiriendo que la inhibici\u00f3n de esta quinasa podr\u00eda estar implicada en la represi\u00f3n que la hormona ejerce sobre la respuesta del promotor a ras oncog\u00e9nico. el antagonismo transcripcional entre ras y tr se observa tambi\u00e9n en otras c\u00e9lulas tumorales de rata, rat\u00f3n y humano y no es un fen\u00f3meno mutuo puesto que el oncog\u00e9n no inhibe sino que potencia la transcripci\u00f3n mediada por t3. En fibroblastos nih3t3, la hormona bloquea la transformaci\u00f3n inducida por ras oncog\u00e9nico cuando se coexpresa con tr. T3 tambi\u00e9n inhibe la transformaci\u00f3n mediada por v-src, un activador de la v\u00eda ras\/mapks.
