En ausencia de estas, la nucleasa hidroliza otras secuencias alternativas. La especificidad mostrada por la enzima para la hidrolisis de los sustratos se puede considerar en cierto modo intermedia entre las de endonucleasas de restriccion y otras nucleasas mas inespecificas como dnasai. La utilizacion de sustratos con bases modificadas quimicamente junto con experimentos de interaccion con el adn han mostrado que la nucleasa se une debil e inespecificamente con ambos surcos de la doble hebra.  mediante experimentos de inmunomicroscopia e inmunodeteccion se demostro una localizacion a nivel de la pared celular para la enzima; asi como la ausencia de la proteina en medios
