{"id":21232,"date":"2018-03-09T09:10:55","date_gmt":"2018-03-09T09:10:55","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/terapia-genica-del-melanoma-vacunas-con-celulas-modificadas-geneticamente\/"},"modified":"2018-03-09T09:10:55","modified_gmt":"2018-03-09T09:10:55","slug":"terapia-genica-del-melanoma-vacunas-con-celulas-modificadas-geneticamente","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/terapia-genica-del-melanoma-vacunas-con-celulas-modificadas-geneticamente\/","title":{"rendered":"Terapia genica del melanoma: vacunas con c\u00e9lulas modificadas gen\u00e9ticamente"},"content":{"rendered":"<h2>Tesis doctoral de <strong> In\u00e9s Moret Tatay <\/strong><\/h2>\n<p>En el presente trabajo de tesis doctoral, se aborda la posibilidad de estimular el sistema inmune mediante la utilizaci\u00f3n de c\u00e9lulas modificadas gen\u00e9ticamente para producir citoquinas, pudiendo mediar en el rechazo de una implantaci\u00f3n de tumor e incrementar la supervivencia de los animales vacunados frente a los controles.  la primera parte del trabajo ha consistido en la caracterizaci\u00f3n de los complejos formados entre los vectores no virales dotap (l\u00edpido) y pei (pol\u00edmero) con el adn, mediante el desarrollo de procedimientos que permitan la cuantificaci\u00f3n del adn que forma parte de los complejos. Posteriormente se ha valorado la estabilidad del adn cuando es sometidos a procesos degradativos.  tras caracterizar dos de los complejos, mayormente utilizados en la terapia g\u00e9nica no viral, se ha aplicado uno de estos complejos en terapias antitumorales in vivo, mediante la vacunaci\u00f3n de los animales (ratones) son c\u00e9lulas tumorales modificadas gen\u00e9ticamente para producir citoquinas y posteriormente irradiadas. se han empleado pl\u00e1smidos que codifican para las citoquinas gm-csf y para la il-4, por ser mediadores del sistema inmune, y como l\u00ednea celular la b16 que corresponden a un melanoma murino.  se ha empezado la vacunaci\u00f3n utilizando de las c\u00e9lulas transfectadas. La administraci\u00f3n in vivo de los clones celulares productores de gm-csf ha resultado en la supervivencia del 100% de los animales vacunados con la dosis m\u00e1s elevada utilizada,y que ha descendido al 80% cuando la dosis se ha reducido de 1 mill\u00f3n a 0.2 millones por rat\u00f3n. Sin embargo la producci\u00f3n de il-4 no ha mostrado efecto antitumoral, ni la administraci\u00f3n conjunta de c\u00e9lulas productoras de gm-csf y de c\u00e9lulas productoras de il-4.  posteriormente se ha utilizado como vacuna c\u00e9lulas transfectadas pero no seleccionadas (transfecci\u00f3n reciente), que tambien ha permitido que el 100% de los ratones sobreviva a una
