{"id":22066,"date":"2018-03-09T09:12:06","date_gmt":"2018-03-09T09:12:06","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/purificacion-caracterizacion-y-mecanismos-adaptativos-de-la-glutamina-sintetasa-en-la-oxifotobacteria-marina-prochlorococcus\/"},"modified":"2018-03-09T09:12:06","modified_gmt":"2018-03-09T09:12:06","slug":"purificacion-caracterizacion-y-mecanismos-adaptativos-de-la-glutamina-sintetasa-en-la-oxifotobacteria-marina-prochlorococcus","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/purificacion-caracterizacion-y-mecanismos-adaptativos-de-la-glutamina-sintetasa-en-la-oxifotobacteria-marina-prochlorococcus\/","title":{"rendered":"Purificaci\u00f3n, caracterizaci\u00f3n y mecanismos adaptativos de la glutamina sintetasa en la oxifotobacteria marina prochlorococcus"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Sabah El Alaoui <\/strong><\/h2>\n<p>Se ha estudiado la regulaci\u00f3n de la glutamina sintestasa (gs) de tres estirpes de prochlorococcus (ss120, med4, pcc9511), analizando los niveles de actividad transferasa (determinaci\u00f3n espectrofotom\u00e9trica del gamma-glutamil hidroxamato formado) y biosint\u00e9tica (determinaci\u00f3n mediante hplc de glutamina producida), as\u00ed como la concentraci\u00f3n intracelular de la prote\u00edna gs (determinada mediante inmunoelectroforesis cuantitativa).  la actividad glutamina sintetasa disminu\u00eda cuando los cultivos se somet\u00edan a ausencia de nitr\u00f3geno o en presencia de fuentes de nitr\u00f3geno no asimilables.  la ausencia de f\u00f3sforo ten\u00eda un efecto a\u00fan m\u00e1s marcado sobre la actividad dela enzima.  el msx provocaba una inactivaci\u00f3n de la enzima menor que en otros organismos fotosint\u00e9ticos; por otro lado, la adici\u00f3n de azaserina provocaba un incremento de la actividad. La oscuridad no afecvta de forma significativa la actividad de la enzima, en tanto que la respuesta antag\u00f3nica de los inhibidores dcmu y dbmib en alguna estirpe parece indicar que las plastoquinonas podr\u00edan estar implicadas en la regulaci\u00f3n de esta enzima.  por otro lado, la cuantificaci\u00f3n de la concentraci\u00f3n de prote\u00edna gs en extractos celulares de cultivos sometidos a diferentes condiciones mostr\u00f3 que la azaserina induc\u00eda un incremento en la concentraci\u00f3n de la enzima; sin embargo, otras condiciones (ayuno de nitr\u00f3geno, adici\u00f3n de inhibidores como msx, dcmu o dbmib) no parecen afectar significativamente la concentraci\u00f3n de la gs.  hemos purificado la glutamina sintetasa de dos estirpes de prochlorococcus (pcc9511 y ss120).  se obtuv\u00f3 una preparaci\u00f3n eletrofor\u00e9ticamente homog\u00e9na. Se calcularon los valores de km para los ensayos de actividad transferasa y biosint\u00e9tica, observ\u00e1ndose valores similares entre ambas estirpes.  el peso molecular estimado fue de 48,6 kda. La temperatura \u00f3ptima para la actividad transferasa es de 55\u00c2\u00bac, el ph \u00f3ptimo de 7,5.
