Tienen una funci\u00f3n protectora esencial para la estabilidad celular pues evitan que los extremos cromos\u00f3micos sean confundios con roturas de dna. La telomerasa es un reversotranscriptasa que sintetiza repeticiones telom\u00e9ricas di novo. La mayor parte de las c\u00e9lulas primarias y de los tejidos som\u00e1ticos carecen de actividades telomerasa de modo que sus tel\u00f3meros se acortan con las divisiones celulares. Por debajo de una longitud m\u00ednima los tel\u00f3meros pierden su funcionalidad dando lugar a una inestabilidad cromos\u00f3mica que compromete la viabilidad celular. Las c\u00e9lulas tumorales poseen altos niveles de actividad telomerasa que les permiten proliferar indefinidamente. Los ratones deficientes en telomersas con tel\u00f3meros cortos muestrasn defectos en tejidos altamente proliferativos como consecuencia de una menor proliferaci\u00f3n celular y una menor tasa de apoptosis. En esta tesis se estudia la susceptibilidad tumoral de este modelo de rat\u00f3n. Los resultados obtenidos demuestran que los tel\u00f3meros cortos act\u00faan como potentes inhibidores tumorales. Adem\u00e1s hacen a las c\u00e9lulas m\u00e1s sensibles al tratamiento con agentes genot\u00f3xicos. Por otra parte, la ausencia de telomerasa incluso en presencia de tel\u00f3meros largos disminuye la incidencia tumoral, sugiriendo la existencia de funciones adicionales de la telomerasa ademas de la elongaci\u00f3n telom\u00e9rica. En esta tesis se describen tambi\u00e9n la obtenci\u00f3n y caracterizaci\u00f3n de un modelo de animales transg\u00e9nicos con altos niveles de expresi\u00f3n de telomerasa en diversos tejidos: los animales k5-mtert. Nuestros resultados muestran que la sobreexpresi\u00f3n de telomerasa no modifica significativamente la longitud telom\u00e9rica ni la estructura de los tejidos a edades tempranas. sin embargo, la presencia de altos niveles de telomerasa favorece la proliferaci\u00f3n celular tras el tratamiento con \u00e9steres de<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
