Isabel Parra Bola <\/strong><\/h2>\n<p>El sistema de activaic\u00f3n del plasmin\u00f3geno es el proceso mediante el cual el zim\u00f3geno inactivo plasmin\u00f3geno es convertivo en su forma activa plasmina por acci\u00f3n de dos proteasas extracelulares que act\u00faan como activadores espec\u00edficos, el activador del plasmin\u00f3geno de tipo uroquinasa o upa y el de tipo tisular o tpa. La generaci\u00f3n de plasmina est\u00e1 regulada negativamente a nivel de los activadores por acci\u00f3n de inhibidores espec\u00edficos de los activadores del plasmin\u00f3geno, o pais, y a nivel de la propia plasmina por la alfa2-antiplasmina.  est\u00e1 descrito que la proteolisis pericelular mediada por plasmina juega un papel importante de la invasi\u00f3n tumoral y la met\u00e1stasis. De hecho, se ha observado una expresi\u00f3n elevada de algunos de los componentes del sistema, como upa y pai-1, en c\u00e9lulas tumorales y\/o en el estroma que las rodea, siendo considerados factores de mal progn\u00f3stico. Este hecho nos ha llevado a plantear si en el contexto de una c\u00e9lula normal, no tumoral, su expresi\u00f3n pudiera estar modulada por el tratamiento con agentes genot\u00f3xicos ambientales, conocidos inductores de carcinog\u00e9nesis.  En esta tesis demostramos, en primer lugar, que la radiaci\u00f3n uv y el agente alquilante mnng inducen la expresi\u00f3n del gen upa a trav\u00e9s de un mecanismo transcripcional, siendo un elemento enhancer de tipo ap1 localizado a -2.4 kb del sitio de inicio de la transcripci\u00f3n del promotor de upa, el responsable de dicha inducci\u00f3n. adem\u00e1s, hemos determinado que la v\u00eda de se\u00f1alizaci\u00f3n de jnk (y no erk o p38) es la responsable de la activaci\u00f3n transcripcional de upa en respuesta a ambos agentes.  en segundo lugar, hemos demostrado que, al igual que upa, su inhibidor fisiol\u00f3gico pai-1 tambi\u00e9n es inducido en respuesta a mnng.
