Para abordar este problema en escherichia coli, construimos varios pl\u00e1smidos con un origen cole1 y una barrera polar para la replicaci\u00f3n localizada a distintas distancias del origen. El an\u00e1lisis de los intermediarios de replicaci\u00f3n de estos pl\u00e1smidos por electroforesis bidimensional en geles de agarosa y microscopia electr\u00f3nica nos ha permitido comprobar que el superenrollado negativo disminuye a medida que avanza la horquilla.  hemos comprobado que la cloroquina y el bromuro de etidio, que inducen el superenrollamiento positivo de mol\u00e9culas no replicadas, son incapaces de inducir en superenrollamiento positivo de los intermediarios de replicaci\u00f3n. esto se debe a que en los intermediarios el superenrollamiento positivo es inmediatamente adsorbido por una regresi\u00f3n de las horquillas. El anudamiento de las doble-h\u00e9lices hermanas dentr\u00e1s de la horquilla ocurre normalmente durante la replicaci\u00f3n.  para estudiar la din\u00e1mica de la formaci\u00f3n de nudos, utilizamos geles bidimensionales para identificar burbujas anudadas en mol\u00e9culas con la horquilla detenida en distintos sitios a lo largo del proceso. Hemos comprabado que el n\u00famero y complejidad de burbujas anudadas aumenta en funci\u00f3n del tama\u00f1o de la burbuja, lo que sugiere que la probabilidad de anudamiento es inversamente proporcional a la densidad de pre-encadenados.  finalmente, estudiamos las consecuencias topol\u00f3gicas de la colisi\u00f3n frontal entre replicaci\u00f3n y trascripci\u00f3n.  los resultados obtenidos indican que este tipo de colisi\u00f3n favorece la formaci\u00f3n de burbujas anudadas y que un exceso de intermediarios anudados tendr\u00eda consecuencias delet\u00e9reas para la segregaci\u00f3n. Esta podr\u00eda ser al menos una de las razones por lo que a lo largo de la evoluci\u00f3n los genomas han evitado la colisi\u00f3n frontal entre transcripci\u00f3n y
