La clonacion del gen (ssg1) que codifica una exo-1,3-beta-glucanasa especifica de la esporulacion, se consiguio mediante el escrutinio de una genoteca de s.  cerevisiae con una sonda de dna amplificada por pcr, empleando oligonucleotidos sinteticos basados en la secuencia de aminoacidos del extremo amino-terminal de la proteina purificada. Ssg1 codifica un polipeptido de 445 aminoacidos (51.8 kda) que presenta una fuerte homolog\u00eda con las exo-beta-glucanasas vegetativas codificadas por los genes exg1 y exg2 de s. Cerevisiae. La region amino-terminal del producto primario del gen es un dominio muy hidrofobico con todas las caracteristicas de los peptidos se\u00f1al de las proteinas de secrecion. El analisis \u00abnorthern\u00bb pone de manifiesto la existencia de un transcrito (1.7 kb) del gen ssg1, que se detecta unicamente en diploides esporulantes (matalfa\/matalfa), pero que no aparece en celulas creciendo vegetativamente ni en diploides no esporulantes (matalfa\/matalfa) incubados en condiciones de privacion de nitrogeno. Ssg1 se induce en los estadios tardios de esporulacion, alcanzando el nivel maximo de expresion durante el periodo de formacion de la pared de las ascosporas.  diploides mutantes ssg1\/ssg1 son capaces de completar el proceso de la esporulacion, aunque muestran un significativo retraso en la aparicion de ascas maduras, lo que indica que el producto de este gen lleva a cabo funciones importantes, aunque no imprescindibles, en la formacion de la pared de las esporas. El gen ssg1 se localiza en el brazo derecho del cromosoma xv, situado a 8.2 cm del gen his3 y estrechamente ligado al gen ade9.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
