Bego\u00f1a <\/strong><\/h2>\n<p>Esta tesis describe la producci\u00f3n de compuestos antitumorales empleando prote\u00ednas inactivadoras de ribosomas (rips), unas enzimas vegetales que inhiben la bios\u00edntesis proteica.  los nuevos agentes terap\u00e9uticos se han dise\u00f1ado para destruir la neovasculatura tumoral- y de este modo, provocar el colpaso del tumor- sin da\u00f1ar a los tejidos sanos. Para ello, los compuestos se han enviado hacia dos ant\u00edgenos espec\u00edficos de las c\u00e9lulas endoteliales de los vasos tumorales: la endoglina humana y la sintegrinas del tipo alfav.  en primer lugar, se han producido en escherichia coli dos rips recombinantes, las musarminas 1 y 4. Su purificaci\u00f3n cromatogr\u00e1fica ha permitido recuperar prote\u00ednas homog\u00e9neas, que mantienen la actividad de sus hom\u00f3logas nativas.  las dos prote\u00ednas anteriores, junto con otras rips monom\u00e9ricas (saporina s6 y momordina i nativas), se han utilizado en la construcci\u00f3n d einmunotoxinas, compuestos formados por la uni\u00f3n qu\u00edmica de una rip y un anticuerpo, que se une selectivamente a una determinada mol\u00e9cula blanco. De los conjugados producidos, 4 se dirigen a la endoglina humana, mientras que el resto reconoce un anticuerpo monoclonal que, a su vez, se fija a la endoglina humana.  las inmunotoxinas generadas han resultado muy eficaces in vitro, ya que en un intervalo amplio de concentraciones eliminan c\u00e9lulas con endoglina sin afectar a las dem\u00e1s c\u00e9lulas. Adem\u00e1s, entre los nuevos preparados se han obtenido, por primera vez, conjugados  anti-endoglina en los que intervienen dos rips recombinantes (musarminas 1 y 4), que son potencialmente menos inmunog\u00e9nicos y t\u00f3xicos que los que contienen rips nativas.  asimismo, se ha creado una prote\u00edna de fusi\u00f3n t\u00f3xica formada por la mu1 y un p\u00e9ptido de adhesi\u00f3n a integrinas alfav.
