La rmn pramagn\u00e9tica era tradicionalmente evitada por la p\u00e9rdida de informaci\u00f3n en las proximidades del entorno met\u00e1lico. En esta tesi sse demuestra, sin embargo, que el paramagnetismo en rmn proporciona datos estructurales (derivados de las velocidades de relajaci\u00f3n, de los desplazamientos de pseudocontacto y de los acoplamientos dipolares residuales) que compensan y superan la p\u00e9rdida de informaci\u00f3n por relajaci\u00f3n.  los sistemas de estudio han sido la prote\u00edna azul de cobre (bcp) rusticianina (rc) y la prote\u00edna activadora de calcio calbindina d9k (cb). Rc es la bcp con el mayor potencial redoz (680 mv), siendo, asimismo, muy estable a valores de ph extremadamente \u00e1cidos. Hemos caracterizado la interacci\u00f3n del metal con sus ligandos mediante estudios de rmn paramagn\u00e9tica tanto del sistema nativo, cu(ii), como del derviado de cobalto(ii). La estructura electr\u00f3nica del ion cobre viene determinada por la interacci\u00f3n de \u00e9ste con los ligandos histidina, mientras que la interacci\u00f3n del cobre con los ligandos ciste\u00edna y metionina es debido al plegamiento global y en las proximidades del entorno met\u00e1lico de la prote\u00edna. La interacci\u00f3n cu-sgammacys es tanto m\u00e1s fuerte cuanto m\u00e1s cerca est\u00e1 el ion met\u00e1lico del plano nhisnhisscys formado por los \u00e1tomos dadores de los ligando ecuatoriales. El estudio de mutantes de rc en la metionina axial revela que aquellos amino\u00e1cidos que proporcionan al cobre una geometr\u00eda m\u00e1s tetra\u00e9drica (mutantes de glutamina, glutamato y ciste\u00edna) desplazan al cobre de ese plano y disminuyen la velocidad de autotransferencia electr\u00f3nica. Mutantes en esta metionina que obligan al cobre a permanecer en dicho plano (leucina) favorecen la velocidad de transferencia electr\u00f3nica.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la
