{"id":28912,"date":"2018-03-09T09:21:48","date_gmt":"2018-03-09T09:21:48","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/transferencia-de-genes-a-protoplastos-de-melon\/"},"modified":"2018-03-09T09:21:48","modified_gmt":"2018-03-09T09:21:48","slug":"transferencia-de-genes-a-protoplastos-de-melon","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/politecnica-de-valencia\/transferencia-de-genes-a-protoplastos-de-melon\/","title":{"rendered":"Transferencia de genes a protoplastos de mel\u00f3n"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Alejandro Atar\u00e9s Huerta <\/strong><\/h2>\n<p>El mel\u00f3n es una de las especies hort\u00edcolas m\u00e1s importantes en el mundo. en este trabajo pretendemos abordar el desarrollo de diversos m\u00e9todos que permitan la transferencia de genes al acervo g\u00e9nico de mel\u00f3n mediante la transformaci\u00f3n y fusi\u00f3n de protoplastos.  en primer lugar, empleando el protocolo de aislamiento, cultivo y regeneraci\u00f3n de plantas a partir de protoplastos de mel\u00f3n, desarrollado previamente en nuestro grupo, estudiamos dos v\u00edas para la selecci\u00f3n de las c\u00e9lulas procedentes de protoplastos transformados o fusionados. Utilizamos cotiledones de ocho d\u00edas con dos d\u00edas de precultivo en medio nb 2,5\/1,0 como fuente de protoplastos y comprobamos que las c\u00e9lulas regeneradas no pod\u00eda seguir su crecimiento si se a\u00f1ad\u00edan 5 mg.L-1 de kanamicina desde el inicio del cultivo o 50 mg.L-1 de kanamicina en la fase de gelificaci\u00f3n. En cambio, en la fase de minicallos, ni siquiera una concentraci\u00f3n de 100 mg.L-1 de kanamicina era suficiente para inhibir el crecimiento. Por otra parte, vimos que la adici\u00f3n desde el inicio del cultivo de nacl 50 nm era suficiente para impedir el crecimiento de los protoplastos de mel\u00f3n. En cambio, cuando se aplicaba el mismo agente selectivo en la fase de gelificaci\u00f3n, hac\u00eda falta un tratamiento con nacl 150 mm para detener el crecimiento, aunque a menores concentraciones tambi\u00e9n ve\u00edamos un claro efecto sobre el cultivo.  conviene resaltar que se ha conseguido la trasformaci\u00f3n estable de nuestro material, mediante dos m\u00e9todos, la electroporaci\u00f3n (transformaci\u00f3n de protoplastos mediante el empelo de corrientes el\u00e9ctricas) y la quimioporaci\u00f3n (transformaci\u00f3n de protoplastos mediante la adici\u00f3n de polietilenglicol). En el primer caso, hemos comprobado la importancia que tiene la adici\u00f3n de calcio en el medio de electroporaci\u00f3n cuando \u00e9sta se lleva a cabo a temperatura ambiente. en ambos
