La exposici\u00f3n de c\u00e9lulas de origen fibrobl\u00e1stico (nih3t3) y nerobl\u00e1stico (c1300) a antiestr\u00f3genos (ae, toremifeno y tamoxifeno) activan el canal maxi-cl- en la configuraci\u00f3n electrofisiol\u00f3gica de c\u00e9lula entera (whole cell), permitiendo as\u00ed el estudio del maxi-cl- en membranas celulares sin escindir. El pretratamiento de las c\u00e9lulas con 17?? Estradiol o ampc previene la activaci\u00f3n por ae delas corrientes del canal maxi-cl-. Este efecto inhibidor fue anulado en presencia de estaurosporina, un inhibidor por prote\u00ednas quinasas que fosrolan residuos del tipo serina\/treonina.  la activaci\u00f3n del canal por ae tambi\u00e9n se puede prevenir por la adici\u00f3n intracelular de \u00e1cido okadaico (ao, 1 nm), una toxina que a esta concentraci\u00f3n inhibe completamente la actividad de la prote\u00edna fosfatasa 2a (pp2a). Sin embargo, la actividad total de la pp2a no cambia en c\u00e9lulas expuestas tanto a estradiol como a tniestr\u00f3genos, por lo que aun no est\u00e1 totalmente esclarecido el proceso mediante el cual la fosfatasa 2a act\u00faa modulando el canal. Nuestra hip\u00f3tesis es que el canal maxi-cl se mantiene cerrado mediante un evento de fosforilaci\u00f3n del canal o de aun prote\u00edna reguladora asociada a este. en presencia del ae; el canal es activado por un proceso de desforforilaci\u00f3n a trav\u00e9s de la acci\u00f3n de una fosfatasa tipo pp2a. Un gran obst\u00e1culo en el estudio de la regulaci\u00f3n por e2 y ae del maxi-cl- era el desconocimiento de la identidad molecular del maxi-cl. Las propiedades electrofisiol\u00f3gicas del maxi-cl- se asemeja a aquellas descritas para el canal ani\u00f3nico vdac, una prote\u00edna integral de membrana que abunda en la membrana externa de la mitocondria. Esta prote\u00edna proporciona la v\u00eda principal para el transporte de diferentes sustratos tales como atp y coleterol y ha sido involucrada<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la
