Dada la importante estimulaci\u00f3n del sistema renina-angiotensina en la hipertensi\u00f3n arterial, la hip\u00f3tesis de este trabajo se centr\u00f3 en el estudio del efecto de la leptina sobre las acciones de la angiotensina (ang) ii sobre la musculatura lisa vascular y la posible participaci\u00f3n del no en este efecto.  en primer lugar, se detect\u00f3 la expresi\u00f3n de arnm del receptor de leptina (ob-r) en c\u00e9lulas de m\u00fasculo liso vascular (cmlvs) de ratas wistar mediante rt-pcr. Los an\u00e1lisis de western-blot e inmunocitoqu\u00edmica confirmaron la expresi\u00f3n proteica de ob-r en cmlvs. A continuaci\u00f3n, se analiz\u00f3 la producci\u00f3n de no en cmlvs estimuladas con leptina (10-8 mol\/l) durante 24 h. La cuantificaci\u00f3n de los no-3 y no-2 en los medios de cultivo mediante el m\u00e9todo de griess mostraron que la leptina induc\u00eda un ligero aumento de la producci\u00f3n de no (p menor 0,05) en las cmlvs desde la primera hora del est\u00edmulo. Tras el est\u00edmulo, las cmlvs fueron lisadas para determinar la actividad de las sintasas de o (nos) mediante la conversi\u00f3n de [3h]arginina en [3h]citrulina. los homogenados de cmlvs estimulados con leptina (10-8 mol\/l) presentaron un aumento de la actividad nos (p menor 0,05) respecto a los controles sin est\u00edmulo. La ausencia de calcio en la reacci\u00f3n no modific\u00f3 este efecto de la leptina (p=0,443). Finalmente, se estudi\u00f3 el efecto de la leptina sobre las acciones vaculares de la ang ii. La leptina (10-8 mol\/l) inhibi\u00f3 el aumento de ca2+ citos\u00f3lido inducido por la ang ii (10-7 mol\/l) .
