Demostramos que los fibroblastos primarios de rat\u00f3n con tel\u00f3meros cr\u00edticamente cortos (g4 terc-\/-), tienen una disfunci\u00f3n telom\u00e9rica similar a la provocada por la deficiencia de ku86. Estas disfunci\u00f3n induce senescencia prematura y evita la inmortalizaci\u00f3n espont\u00e1nea de los fibroblastos.  con el an\u00e1lisis de diferentes fondos gen\u00e9ticos de ratones terc-\/- deficientes a su vez, ya sea en ku86, dna-pkcs o parp-1 concluimos que ku86 y dna-pkcs median las fusiones extremo-extremo originadas por los tel\u00f3meros cr\u00edticamente cortos y que la ausencia de ambas prote\u00ednas no afecta la longitud del g-strand overhang. As\u00ed mismo, demostramos que ku86 es un regulador negativo de la telomerasa, mientras que dna-pkcs participa con \u00e9sta para mantener los tel\u00f3meros. Confirmamos, por otra parte, que parp-1 no participa en el mantenimiento del tel\u00f3mero, ni afecta su funci\u00f3n.  analizamos el fenotipo de ratones deficientes en dna-pkcs-\/- y vimos que, comaprados con los wild-type, estos ratones tinenen una longevidad menor, envejecen prematuramente, son m\u00e1s peque\u00f1os y son m\u00e1s susceptibles de desarrollar linfomas de timo y atrofia intestinal.  el estudio detallado del fenotipo de los ratones terc-\/- simult\u00e1neamente deficientes en ku86, dna-pkcs o parp-1 demostr\u00f3 que ku86 y dna-pkcs aceleran la tasa de mortalidad y los defectos proliferativos provocados por el acortamiento progresivo de los tel\u00f3meros. As\u00ed mismo encontramos un sinergismo entre el acortamiento telom\u00e9rico progresivo y la p\u00e9rdida de ku86, dna-pkcs, y en menor grado parp-1, en el envejecimiento prematuro.
