La transcripcion desde el promotor del mmtv disminuye progresivamente durante el tratamiento hormonal, cinco dias, mientras que la de los otros genes regulados por glucocorticoides se mantiene estable. Este fenomeno se observa en diferentes lineas celulares. La desactivacion del promotor del mmtv se correlaciona temporalmente con una disminucion en el contenido nuclear del receptor de glucocorticoides (gr) y con una disminucion concomitante en la hipersensibilidad a la dnasal, que se interpreta como un cierre en la estructura de la cromatina del promotor. Asi mismo se ha observado que las formas del promotor mmtv transfectadas transitoriamente mantienen estable su expresion durante todo el tiempo del tratamiento con hormona. La menor afinidad del promotor mmtv por el gr que hemos observado podria explicar su desactivacion en un contexto de perdida de gr pues un menor nivel de gr no seria suficiente para mantener abierta su cromatina y si bastaria para mantener activos los demas genes hormono-regulados. La particular estructura de la cromatina del promotor mmtv integrado en el genoma de las celulas y su inactividad en ausencia de hormona sugieren que el factor que determina los requerimientos de gr
