En la presente tesis \u00abaplicabilidad de los metodos de amplificacion genetica en el diagnostico de la tuberculosis\u00bb se han estudiado y optimizado las condiciones de pretratamiento de las muestras clinicas para su posterior uso en la extraccion, amplificacion y deteccion de dna y rna micobacteriano, buscando tambien muy especificamente la neutralizacion de los elementos inhibidores de los procesos de amplificacion genetica. Ademas fueron evaluados diferentes metodos de amplificacion de dna y rna que permiten la deteccion directa de los complejos m.  tuberculosis y m.Avium-m.Intracellulare en diferentes tipos de muestras clinicas, comparando los resultados obtenidos en los metodos moleculares con los obtenidos con las tecnicas de examen microscopico y cultivo en medios solidos convencionales y en el sistema bactec. En este trabajo se demuestra que concentraciones de lauril sulfato de sodio (sds) superiores a 3.2 mg\/ml inhiben las reacciones de amplificacion de rnar en la deteccion de m.  tuberculosis directamente de muestras clinicas descontaminadas con la tecnica de tacquet y tisson. El posterior lavado de las muestras clinicas con 30 ml de agua destilada esteril elimina el sds a niveles suficientes como para no interferir en las reacciones de amplificacion. Realizando este procedimiento (descontaminacion con tacquet-tisson y posterior lavado con 30 ml de agua destilada) a las muestras clinicas, decidimos evaluar la aplicabilidad de las tecnicas de amplificacion genetica en el diagnostico de la tuberculosis pulmonar y extrapulmonar. La tecnica de amplificacion de rna (amplified mycobacterium tuberculosis direct test; amtdt) es una tecnica con una buena sensibilidad (80 y 90%, respectivamente, en muestras extrarrespiratorias y
