La actividad determinada ex-vivo relaciona factores farmacocineticos, microbiologicos i farmacodinamicos.  estudio en fase 1 a dosis unica de rufloxacina (ruf) (400 mg.) Y norfloxacina (nor) (400 mg.), Via oral aleatorizado y con dise\u00f1o cruzado, en el que participaron 12 voluntarios sanos.  se determinaron los niveles urinarios y plasmaticos por metodo microbiologico y hplc, y niveles en heces por metodo microbiologico. Se determino la actividad bactericida urinaria frente a cepas control y tambien la capacidad predictiva de la actividad teorica frente a la experimental.  resultados: las concentraciones urinarias de nor fueron altas en los periodos iniciales. La ruf mantiene niveles estables las primeras 48 horas. En los parametros cineticos, el tiempo medio de residencia es superior para ruf (p 0,01), por hplc tambien lo son la auc 0-72 y el tmax.  las concentraciones plasmaticas de ruf son superiores a las de nor por ambos metodos. Los valores de ruf (hplc) de cmax (4,35 g\/ml), tl\/2 (27,6 h.) Y auc 0- (157,4 g.H\/ml) son superiores a los de nor (p 0,01).  por el metodo microbiologico las concentraciones en heces a las 48 horas de nor fueron superiores a las de ruf (p 0,01). A las 168 horas se determino ruf en 9 voluntarios y solo en 1 de nor (p 0,01).  se determinaron los poderes bactericidas urinarios experimentales y teoricos y las auc de los poderes.  para la ruf la razon experimental\/teorica fue siempre superior a 1, ya que los valores experimentales fueron superiores a los esperados en las dos cepas.  conclusiones: se determinan para la nor unos niveles mucho mas elevados en los periodos iniciales, cayendo rapidamente.
