Ha sido desarrollado un metodo de obtencion de protoplastos con propiedades funcionales mejoradas, mediante la casi eliminacion de la actividad proteasa asociada al complejo litico empleado.  han sido ensayadas diferentes estrategias de carga de indicadores fluorescentes de calcio (sintesis de nuevos derivados lipofilicos, y metodos de carga acida y facilitada por digitonina). Estos metodos se han demostrado inapropiados.  ha sido desarrollado un sistema de microscopia de fluorescencia asociada a tecnicas de digitalizacion y analisis de imagenes por ordenador, para la obtencion de mapas intracelulares de concentraciones de iones (calcio y ph).  ha sido medido el ph citosolico de penicillium cyclopium y se ha determinado que el calcio causa una rapida alcalinizacion citosolica, asi como una disipacion del gradiente de ph citosolico presente en hifas en crecimiento vegetativo (la zona apical mas acida). La alcalinizacion citosolica, que es necesaria para el desarrollo de la conidiacion, no parece mediada ni por una activacion estable de la h+-atpasa de membrana plasmatica ni por una alteracion en la permeabilidad de la membrana a los h+. Bloqueadores de canales de calcio son inefectivos para inhibir la conidiacion.  el calcio se une a la membrana plasmatica del con una afinidad ligeramente superior que el magnesio (cation no inductor). Esto sugiere un efecto especifico del calcio, posiblemente por interaccion con alguna proteina de la membrana plasmatica.  el calcio origina una disminucion en el grado de fosforilacion de una proteina de 34 kda de la fraccion microsomal de membrana.  todos los resultados sugieren que el efecto inicial del calcio ocurre en la superficie externa de la membrana plasmatica.
