Jes\u00fas <\/strong><\/h2>\n<p>La produccion y el metabolismo del precursor de la proteina beta-amiloide tienen gran importancia en la patogenia de la enfermedad de alzheimer. Se ha estudiado la regulacion de la expresion del gen de app en las celulas pc 12, un feocromocitoma de rata, y en las celulas procedentes de neuroblastomas sh sy5y y n2a, todas ellas con capacidad de adquirir un fenotipo neuronal ante ciertos estimulos. El tratamiento de la linea celular pc12 con factores de crecimiento que actuan a traves de receptores con actividad tirosina quinasa como el ngf y con forskolina induce la expresion del gen de app mediante un mecanismo de regulacion transcripcional donde las secuencias mediadoras del efecto inductor de estos agentes se encuentra se localizan en el mismo fragmento de 117 pb del gen app.  asimismo, la regulacion de app por ngf requiere la presencia de ras funcional en la celula. Se ha observado que en la linea celular sh sy5y tanto el ester de forbol tpa como el acido retinoico son capaces de inducir transcripcionalmente la expresion del gen de app a tiempos y de manera especifica de linea celular.  adicionalmente se ha analizado el efecto represor de la hormona tiroidea sobre la expresion del precursor de la proteina beta-amiloide en las celulas n2a. Esta regulacion transcripcional negativa por t3 no es especifica del tipo de receptor para hormonas tiroideas y es mediada por las mismas secuencias de dna que median la respuesta a ngf y fk en las celulas pc12. Ademas, t3 es capaz de modificar el procesamiento alternativo de las isoformas mayoritarias de 770 y 695 aminoacidos.
