La aplicabilidad clinica de la cya esta sin embargo, limitada por sus efectos secundarios de caracter no inmunologico; los mas graves son su caracter hepatotoxico y nefrotoxico. Los objetivos generales de este trabajo fueron: 1) estudio del da\u00f1o renal provocado por la cya en un modelo experimental de nefrotoxicidad aguda, su relacion con la dosis suministrada y el tiempo de tratamiento. 2) estudio de mecanismos capaces de explicar esa nefrotoxicidad. Se encontro una correlacion entre el da\u00f1o renal y la sintesis de sustancias vasoconstrictivas (tromboxano a2), radicales libres (ros), niveles de peroxidacion lipidica (malonildialdehido) y activacion de la defensa antioxidante enzimatica (superoxido dismutasa, catalasa y glutation peroxidasa). Se establecio una hipotesis acerca de que el origen del da\u00f1o renal estaba en la sintesis aumentada de ros y se intento justificar mediante una aproximacion indirecta: se suplemento exogenamente con el antioxidante vit. E, scavenger de radicales libres, y se estudio la influencia que esta suplementacion tenia sobre los parametros de funcionalidad renal, la sintesis de tx a2, ros y mda, asi como sobre la activacion de la defensa antioxidante enzimatica. Los resultados muestran una reversion de los valores de los parametros de funcionalidad renal a sus cifras basales y una normalizacion de la sintesis del resto de parametros medidos. Proponemos la hipotesis de que el da\u00f1o renal observado en el tratamiento con cya esta mediado por una sintesis aumentada de ros y que el aumento de las defensas antioxidantes por suplementacion
