Hemos analizado la expresion dle gen de pal-1 en celulas endoteliales de cordon umbilical humano (huvec) utilizando northern blot y transcripcion inversa-reaccion en cadena de la polimerasa (rt-pcr) con primers biotinilados. La concentracion de pai-1 antigenico se determino en los sobrenadantes mediante tecnica de elisa.  los cultivos se estimularon con lps (50ng\/ml), tnfalfa (500 u\/ ml) e il-1 alfa (500 u\/ml), hnf (1-10 u\/ml) hbmp(1-10 u\/ml) e hirudina (20-100 u\/ml). Se recogieron las celulas y los sobrenadantes de los cultivos, antes de la estimulacion, a las 2, 6 y 24 horas. El tratamiento de huvec con endotoxina, tnf e il-1 incremento los niveles de mrna del pai-1. En todos los casos se observo un efecto maximo con la il-1 a las 6 h (12 veces el valor basal). El pai-1 antigenico aumento significativamente a las 24 h de la estimulacion, especialmente en los cultivos estimulados con il-1. No se observo ningun efecto sobre la expresion del pai-1 con las dosis terapeuticas de hnf, hbpm e hirudina. Sin embargo, dosis altas de hnf y hbpm redujeron la expresion del pai-1 y ello correlaciono con un descenso en la secrecion proteica del inhibidor. Estos datos indican que endotoxina y citoquinas inducen un aumento en la expresion endotelial de pai-1, que podria contribuir a la aparicion de trombosis in vivo. Entre los anticoagulantes, unicamente la hnf redujo significativamente la concentracion de mrna del pai-1 en huvec, indicando una accion profibrinolitica.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
