La infeccion cronica por virus c se ha asociado a un estado de depleccion sistemica de este tiol, habiendose sugerido que un aumento del stress oxidativo puede ser la causa del da\u00f1o en la hepatitis cronica por virus c(hcc), objetivos: hemos estudiado la capacidad de sintesis de gsh en el citosol purificado aislado de cmp y la expresion del rna m del gen de la y-glutamilcisteina sintetasa (ggcs), el enzima limitante para su sintesis, en 35 controles (18 mujeres, 17 varones, edad media=37,5 a\u00f1os; rango=20-67) y en 29 pacientes (8 mujeres, 21 varones: edad media=47,8; rango=23-67) con hepatitis cronica activa por virus c (hcc). Materiales y metodos: tras aislar las cmp se purifico su citosol por utlracentrifugacion y se cuantifico la cantidad total de gsh mediante la tecnica de tietze. El citosol se concentro por ultrafiltracion y se dializo la muestra para eliminar su contenido en glutation. Posteriormente se determino la velocidad de sintesis de gsh del citosol dializado cuantificado la formacion del tiol en presencia de sus precursores (cisteina 0,5mm, glutamato 10mm, glicina 10mm), por la fluorescencia a emitida al unirse a monoclorobimano (0,1mm) en presencia de 0,75u\/ml de gsh transferasa. Se considero que la capacidad de sintesis estaba determinada por la diferencia entre la fluorescencia emitida en presencia y ausencia de bso (20mm), un conocido inhibidor de la sintesis de gsh. Asimismo se cuantifico en las mismas muestras el nivel de rnam del enzima ggcs realizando transcripcion reversa y pcr semicuantiativo.  el gen de la b-actina fue utilizado como control interno.  resultados: el gsh total medido en el citosol de los pacientes fue discretamente inferior de los controles (51, 9+1,9nmol\/mg prot vs 46,5 + 2,2), sin que encontramos diferencias significativas entre ambos grupos.
