Estos animales presentan resistencia a la insulina, con hiperinsulinemia, glucemia normal o ligera hiperglucemia, hiperlipidemia, y una tolerancia a la glucosa anormal. La obesidad de estos animales comparte muchas similitudes con la obesidad observada en humanos que se acompa\u00f1a de resistencia a insulina o niddm. En estas situaciones, se ha demostrado que la producci\u00f3n de glucosa hep\u00e1tica es menos sensible a la supresi\u00f3n por insulina y glucosa. En este trabajo hemos estudiado la modulaci\u00f3n por insulina y por glucosa de la gluconeog\u00e9nesis, la cual juega un papel fundamental en la producci\u00f3n de glucosa hep\u00e1tica, en hepatocitos aislados de ratas (fa\/fa). Tambi\u00e9n estudiamos la modulaci\u00f3n de la v\u00eda gluconeog\u00e9nica por sulfonilureas, as\u00ed como su mecanismo de acci\u00f3n, en hepatocitos de estos animales. Estos agentes son hipoglucemiantes orales ampliamente usados en el tratamiento de la niddm, que se sabe inhiben la gluconeog\u00e9nesis hep\u00e1tica por un mecanismo relacionado con el aumento en los niveles de hexosas 6-fosfato y f-2,6-p2, producido por la activaci\u00f3n de la glucogenolisis por dichos agentes.  nuestros resultados muestran que los hepatocitos aislados de ratas (fa\/fa) son resistentes a las acciones a corto plazo de la insulina, glucosa y sulfonilureas sobre la v\u00eda glucol\u00edtica\/gluconeog\u00e9nica. Teniendo en cuenta estos resultados, sorprende el hecho de que los hepatocitos de estos animales tengan m\u00e1s gk, pfk-1 y pk, mayor contenido de f-2,6-p2 y menor actividad pepck, lo que indicar\u00eda que s\u00ed hay una respuesta a la hiperinsulinemia in vivo.  nosotros postulamos que posiblemente en el h\u00edgado de la rata zucker (fa\/fa) se haya producido una adaptaci\u00f3n de la v\u00eda glucol\u00edtica\/gluconeog\u00e9nica a la hiperinsulinemia presente en estos animales, en condiciones basales.
