Uno de los reguladores de este potente sistema es la glicoprote\u00edna plasm\u00e1tica c4b-binding protein (c4bbp). C4bp humano es un heterolig\u00f3mero compuesto por dos subunidades diferentes, c4bpalfa y c4bpbeta, que se organizan en tres complejos de alto peso molecular: alfa7b1, alfa7b0 y alfa6b1. Los polip\u00e9ptidos c4bpalfa y c4bpbeta est\u00e1n codificados por los genes c4bpa y c4bpb, respectivamente, que se expresan en el h\u00edgado. En las cadenas c4bpalfa reside la funci\u00f3n reguladora del complemento, mientras que con la cadena c4bpbeta une la prote\u00edna anticoagulatoria, prote\u00edna s. La uni\u00f3n de la prote\u00edna s a c4bpbeta inactiva su funci\u00f3n cofactora de la prote\u00edna c: puesto que, s\u00f3lo el exceso molar de la prote\u00edna s sobre los polip\u00e9ptidos c4bpbeta se encuentra en forma libre y activa, in vivo debe existir una estrecha regulaci\u00f3n de las isoformas de c4bp que contienen cadena beta, y as\u00ed evitar la interferencia con el normal funcionamiento de la v\u00eda anticoagulatoria de la prote\u00edna c activada. Los resultados obtenidos en esta tesis revelan que son los niveles relativos de expresi\u00f3n de los genes c4bpa y c4bpb los que determinan el patr\u00f3n de isoformas de c4bp. El patr\u00f3n de isoformas de c4bp es adem\u00e1s, heterog\u00e9neo en la poblaci\u00f3n normal. Este patr\u00f3n est\u00e1 determinado gen\u00e9ticamente, es caracter\u00edstico de cada individuo y en condiciones normales permanece constante en el tiempo. Sin embargo, durante la respuesta de fase aguda se produce una elevaci\u00f3n en los niveles plasm\u00e1ticos de c4bp de hasta 4 veces, produci\u00e9ndose cambios en el patr\u00f3n de isoformas de c4bp. Estudios in vitro muestran que estos cambios con consecuencia del efecto diferencial que las citoquinas de fase aguda tienen sobre los genes c4bpa y c4bpb.
