El gen cot est\u00e1 muy relacionado con tpl-2, siendo probablemente su hom\u00f3logo humano. Se ha observado que la forma truncada de cot posee una actividad transformante m\u00e1s elevada que la forma no truncada. En este trabajo demostramos que existen tres mrnas para el gen de cot (cot1, cot2 y cot3). Adem\u00e1s, la transcripci\u00f3n de cot se halla controlada por dos promotores (p1 y p2), de manera p1 controla la expresi\u00f3n de cot1 y cot2 y p2 controla la expresi\u00f3n de cot3. Mientras p2 posee una elevada actividad constitutiva, p2 es fuertemente inducible por est\u00edmulos que activan la respuesta linfocitaria en c\u00e9lulas t. En este trabajo demostramos tambi\u00e9n la existencia de un nuevo ex\u00f3n (3a) que no hab\u00eda sido previamente definido en la estructura gen\u00f3mica del gen cot. Por otra parte, un incremento en la expresi\u00f3n del gen de cot quinasa truncada se correlaciona con un incremento en la producci\u00f3n de interleuquina-2 en c\u00e9lulas jurkat tratadas con ant-cd3. La expresi\u00f3n de cot truncada tambi\u00e9n coopera con pha o pdbu y un ion\u00f3foro de calcio (a23187) en la producci\u00f3n de il-2 en c\u00e9lulas jurkat.  tanto la forma truncada con la no truncada incrementan la transcripci\u00f3n de il-2 ya que aumentan la acumulaci\u00f3n de un gen indicador unido al promotor de il2. La expresi\u00f3n de una forma dominante negativa de cot inhibe la transcripci\u00f3n dirigida por el promotor de il2 en c\u00e9lulas jurkat estimuladas con pdbu y a23187. Nuestros resultados indican que cot quinasa posee un papel en la actividad nuclear de nfat y ap1 en c\u00e9lulas t.
