Este abordaje experimental permite la detecci\u00f3n de todos los tipos de rna del virus: mrna crna y vrna.  utilizando este sistema se ha demostrado que la subunidad pb2 de la rna polimerasa del virus de la gripe se requiere para la replicaci\u00f3n del rna viral, tanto en el primer paso (s\u00edntesis de crna a partir de un vrna molde) como en el segundo (s\u00edntesis de vrna a partir de un crna molde).  se ha reconstitu\u00eddo un sistema in vitro que es capaz de generar crna y mrna virales a partir de extractos nucleares de c\u00e9lulas transfectadas con los genes virales clonados. Al igual que en el sistema in vivo, en este sistema la subunidad pb2 es necesaria para la replicaci\u00f3n del rna viral.  el an\u00e1lisis mutacional de la subunidad pb2 ha confirmado que el determinante principal de la nls de pb2 se localiza en el extremo c-terminal de la prote\u00edna y que adem\u00e1s se requieren otras secuencias internas para que tenga lugar un transporte eficiente. Adem\u00e1s se ha definido la regi\u00f3n n terminal de pb2 como necesaria y suficiente para la interacci\u00f3n con el complejo de la polimerasa. Algunas mutaciones en diversas posiciones de la mol\u00e9cula afectan a la localizaci\u00f3n nuclear de la prote\u00edna, otras alteran su interacci\u00f3n con el complejo transcripcional y otras inhiben su actividad biol\u00f3gica.  de inter\u00e9s especial son los mutantes de inserci\u00f3n que muestran un fenotipo ts o son defectivos en actividad de uni\u00f3n a
