Con la intenci\u00f3n de esclarecer su funci\u00f3n, llevamos a cabo la caracterizaci\u00f3n de componentes celulares que en linfocitos de bazo de rat\u00f3n en proliferaci\u00f3n muestran afinidad por la protalfa enlazada a sepharosa. En esta investigaci\u00f3n encontramos que la mayor parte de los blancos espec\u00edficos de la protalfa en estas c\u00e9lulas son citos\u00f3licos y la actividad proliferativa de las c\u00e9lulas incrementa la concentraci\u00f3n de estos componentes pero no su n\u00famero, de 11 prote\u00ednas principales. Similares componentes con afinidad por protalfa, en movilidad electrofor\u00e9tica, se encontraron en linfocitos tumorales (c\u00e9lulas nc-37), si bien es mayor la presencia, entre \u00e9stos, de prote\u00ednas nucleares.  entre las prote\u00ednas presentes en los subfraccionados de linfocitos de bazo de rat\u00f3n en proliferaci\u00f3n, que presentan afinidad por la protalfa, hemos caracterizado las histonas h3, h2b, h2a y h4, confirmando los datos previos de nuestro laboratorio que han demostrado la interacci\u00f3n de estas prote\u00ednas con la protalfa. Para profundizar en esta afinidad hemos investigado la interacci\u00f3n de la protalfa con los nucleosomas purificados de linfocitos de bazo de rat\u00f3n, mediante experimentos de inducci\u00f3n de enlaces cruzados con el agente qu\u00edmico dsp demostrando que la protalfa se une a los nucleosomas por medio de sus cuatro histonas, principalmente de las h3, h2a y h2b, siendo m\u00e1s destacada esta uni\u00f3n a relaciones molares nucleosomas:protalfa 0,6:1. Cuando las histonas se encuentran libres del nucleosoma, se unen a la protalfa principalmente las histonas h2a y h2b, siendo la relaci\u00f3n molar \u00f3ptima la 0,1:1 (mezcla equimolecular de histonas:protalfa).
