Por experimentos de transfeccion transitoria se ha demostrado que la region minima del promotor de tpo de rata que contiene expresion tiroides-especifica tambien contiene respuesta a tiropropina (tsh) e insulina\/factor de crecimiento similar a insulina (lgf-i) tsh a traves de la via del adenosin monofosfato (ampc) e insulina a traves de los receptores de igf-i inducen la actividad del promotor. La via de la fosfolipasa c (plc) tambien esta involucrada en la regulacion de la actividad del promotor de tpo, ya que el ionoforo de calcio (a23187) y el 12-o tetradecanoil-forbol-13-acetato (tpa) la inhiben rapidamente. Estos datos indican que la region del promotor de tpo utilizada contiene las se\u00f1ales necesarias para su regulacion hormonal. La union del factor nuclear especifico de tiroides 2 (ttf-2) al elemento z del promotor es inducida por tsh forskoina.  dicha union tambien es inhibida por el ionoforo a23187 y por tpa. La actividad de construcciones heterologas que contienen 4, 8 o 12 repeticiones del elemento z del promotor de tpo se induce en respuesta a tsh, forskolina e insulina y se inhibe en respuesta a a23187 o tpa. El mecanismo por el que tsh e insulina\/igf-i inducen la union de tif-2 a dna es por modificacion del factor y no por induccion de su sintesis. Para que un solo sitio de union a ttf-2 actue como un elemento de respuesta a hormonas, como ocurre en el promotor de tpo, se necesita la localizacion precisa de la region b de dicho promotor, a la que se unen los factores de transcripcion ttf-1 y ufb. Ttf-1 debe desempe\u00f1ar algun papel en la regulacion de la transcripcion del gen tpo ya que amplifica tanto la transcripcion basal como el efecto regulador del elemento z.
