Teresa <\/strong><\/h2>\n<p>Esta tesis doctoral se ha centrado en la caracterizaci\u00f3n del sistema de regulaci\u00f3n de la virulencia de l.  monocytogenes, mediado por el activador transcripcional prfa. En primer lugar, determinamos los niveles basales de expresi\u00f3n de los factores de virulencia de l.  monocytogenes in vitro, lo que nos permiti\u00f3 definir por primera vez el fenotipo silvestre de esta especie en lo que se refiere a la producci\u00f3n de factores de virulencia, as\u00ed como identificar y caracterizar mutantes que sobreexpresan constitutivamente dichos factores.  identificamos el origen gen\u00e9tico del fenotipo variante de sobreexpresi\u00f3n de los factores de virulencia, que consiste en el cambio de amino\u00e1cido glyi45ser en el regulador prfa. Esta sustituci\u00f3n bloquear\u00eda a la prote\u00edna reguladora en una conformaci\u00f3n constitutivamente activa con independencia de la presencia o no de un cofactor todav\u00eda no identificado, que ser\u00eda necesario para activar a prfa en las cepas silvestres. Tales observaciones nos han permitido establecer un modelo coherente que explica la regulaci\u00f3n prfa-dependiente.
