De esta manera, para obtener enzimas de elevada pureza a partir de un caldo de fermentaci\u00f3n industrial, se emplean fundamentalmente las distintas t\u00e9cnicas cromatogr\u00e1ficas existentes, siendo el intercambio i\u00f3nico la que ofrece mayores ventajas. Debido a que los costes de operaci\u00f3n de estas unidades de proceso representan la mayor parte del coste total de producci\u00f3n, es necesario llevar a cabo un dise\u00f1o \u00f3ptimo de las etapas cromatogr\u00e1ficas presentes en el proceso global. La presente memoria de investigaci\u00f3n ha sido desarrollada abordando primeramente los conceptos te\u00f3ricos necesarios para ofrecer una visi\u00f3n general acerca de las caracter\u00edsticas principales de la enzima industrial denominada glucoamilasa, as\u00ed como de los procesos existentes para efectuar una l\u00ednea de purificaci\u00f3n de bioproductos, con el fin de ofrecer una introducci\u00f3n al posterior estudio de adsorci\u00f3n por intercambio i\u00f3nico de las dos isoenzimas que forman la glucoamilasa de aspergillus niger. Para realizar este estudio experimental, se ha elegido la resina i\u00f3nica deae-toyopearl 650 por ser la m\u00e1s adecuada, y se han optimizado las condiciones de operaci\u00f3n sobre las que se ha desarrollado posteriormente un trabajo emp\u00edrico secuencial que ha hecho uso de la cromatograf\u00eda de impulso en columnas de hplc, adsorci\u00f3n en tanque agitado discontinuo, y adsorci\u00f3n en continuo en sistemas de lecho fijo. Los resultados as\u00ed obtenidos han sido reproducidos satisfactoriamente mediante los modelos matem\u00e1ticos propuestos para este f\u00edn, obteniendo de esta manera los par\u00e1metros fisicoqu\u00edmicos que rigen este proceso de purificaci\u00f3n determinado. Por \u00faltimo, se ha simulado la operaci\u00f3n de intercambio i\u00f3nico del sistema glucoamilasa-deae-toyopearl 650 a dos escalas superiores a los sistemas empleados en el estudio
