{"id":34869,"date":"1998-01-01T00:00:00","date_gmt":"1998-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-de-chlamydia-psittacii-mediante-inmunomicroscopia-electronica\/"},"modified":"1998-01-01T00:00:00","modified_gmt":"1998-01-01T00:00:00","slug":"estudio-de-chlamydia-psittacii-mediante-inmunomicroscopia-electronica","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-de-la-vida\/estudio-de-chlamydia-psittacii-mediante-inmunomicroscopia-electronica\/","title":{"rendered":"Estudio de chlamydia psittacii mediante inmunomicroscopia electronica."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Antonio  Julian Buendia Marin <\/strong><\/h2>\n<p>En el presente trabajo se ha investigado la localizaci\u00f3n de una serie de ant\u00edgenos de las especies del g\u00e9nero chlamydia que afectan a los rumiantes (chlamydia psittaci serotipo 1 y chlamydia pecorum serotipo 2), utilizando de forma combinada tres tipos de t\u00e9cnicas para inmunomicroscop\u00eda electr\u00f3nica: t\u00e9cnica sin inclusi\u00f3n y t\u00e9cnica pre-inclusi\u00f3n, ambas para el estudio de ant\u00edgenos situados en la superficie clamidial, y t\u00e9cnica post-inclusi\u00f3n que permite la detecci\u00f3n tambi\u00e9n de ant\u00edgenos situados en el interior de la bacteria. Adem\u00e1s utilizando las t\u00e9cnicas puestas a punto en el estudio sobre las clamidias, se ha extendido el trabajo a la localizaci\u00f3n de prote\u00ednas clamidiales expresadas en escherichia coli recombinantes y a la detecci\u00f3n de las clamidias en un modelo de infecci\u00f3n in vivo en ratonas gestantes.  los resultados obtenidos con los ant\u00edgenos de c. Psittaci estudiados muestran que el lipopolisac\u00e1rido clamidial se encuentra localizado en la superficie de los cuerpos reticulares (formas metab\u00f3licas), mientras que en los cuerpos elementales (formas infectivas) su localizaci\u00f3n es interna. Una localizaci\u00f3n similar es encontrada en la prote\u00edna de 80-90 kda (prote\u00edna propuesta como prote\u00edna de diagn\u00f3stico), encontr\u00e1ndose en la superficie de los cuerpos reticulares, y en el espacio peripl\u00e1smico de los cuerpos elementales. Por el contrario, la prote\u00edna de 110 kda (prote\u00edna propuesta para su empleo como vacuna subcelular) y la prote\u00edna de 30 kda se encuentran localizadas en la superficie de ambas formas clamidiales, encontr\u00e1ndose en mayor cantidad la primera. De los diferentes ant\u00edgenos de c. Pecorum testados, tan s\u00f3lo con el lipopolisac\u00e1rido hemos encontrado resultados positivos, siendo estos id\u00e9nticos a los obtenidos con el lipopolisac\u00e1rido de c. Psittaci. El marcaje de la prote\u00edna de diagn\u00f3stico clonada en los e.
