As\u00ed, en tumores mamarios cuyo crecimiento es dependiente de estr\u00f3genos esta hormona presenta efectos antiestrog\u00e9nicos. El objetivo general de este trabajo ha sido determinar el mecanismo por el que esta hormona ejerce estas acciones antiestrog\u00e9nicas, utilizando como sistema experimental las c\u00e9lulas mcf-7. Receptor de estr\u00f3geno-positivas, que derivan de un carcinoma de mama humano. En estas c\u00e9lulas hemos observado que la melatonina inhibe el efecto mitog\u00e9nico de los estr\u00f3genos, asi como la expresi\u00f3n g\u00e9nica controlada por un elemento de respuesta a estr\u00f3genos. Por ello hemos estudiado de que modo la melatonina est\u00e1 interfiriendo con la v\u00eda de se\u00f1alizaci\u00f3n mediada por el receptor nuclear de estr\u00f3genos. Nuestros resultados indican que este efecto inhibidor no se debe a que la hormona pineal impida la uni\u00f3n del estradiol al receptor de estr\u00f3genos, la fosforilaci\u00f3n del receptor o la localizaci\u00f3n nuclear del mismo. Sin embargo la melatonina est\u00e1 inhibiendo la uni\u00f3n del receptor de estr\u00f3genos al elemento de respuesta a estr\u00f3genos, produciendo una inestabilizaci\u00f3n de la uni\u00f3n del receptor al dna.
