Esto conduce a una inhibici\u00f3n general de la traducci\u00f3n y a un incremento en la traducci\u00f3n de un arnm espec\u00edfico que codifica para el factor transcripcional gcn4p, cuya traducci\u00f3n depende de cuatro uorfs presentes en su secuencia leader y de varios factores reguladores en trans gcnp y gcdp. La mayor\u00eda de los factores gcd caracterizados tienen una funci\u00f3n esencial en el inicio de la s\u00edntesis de prote\u00ednas.  gcd13p y gcd15p se identificaron como nuevos represores de la traducci\u00f3n de gcn4. El an\u00e1lisis de los perfiles de polisomas de los mutantes gcd13 y gcd15 muestra un defecto general en la iniciaci\u00f3n de la traducci\u00f3n y la combinaci\u00f3n de las mutaciones gcd15 y gcd13 conduce a la letalidad sint\u00e9tica. Adem\u00e1s, las mutaciones gcd13 y gcd15 reducen la transcripci\u00f3n de varios arnt con mayor efecto sobre la s\u00edntesis del arntj met. Gcd15 y gcd13 codifican las dos subunidades mayores de la arn polimerasa iii en levaduras. La disminuci\u00f3n de la expresi\u00f3n del arntj met probablemente conduce a reducidos niveles del complejo ternario, explicando los defectos en la iniciaci\u00f3n de la traducci\u00f3n general y la desrepresi\u00f3n de la traducci\u00f3n de gcn4 de estos mutantes.  en la segunda parte de la memoria se muestra un estudio detallado del efecto de las mutaciones en gcd14 y la combinaci\u00f3n de \u00e9stas con mutaciones en gcd10 y con la deleci\u00f3n de lhp1. Los mutantes gcd14 acumulan altos niveles de pre-arntj met. La letalidad de la deleci\u00f3n de gcd14 es suprimida por alta dosis de uno de los arntj met, indicando que la funci\u00f3n esencial de gcd14p se requiere para la biog\u00e9nesis del arntj met.
