El genoma se replica una vez por ciclo comenzando la s\u00edntesis de dna en los or\u00edgenes de replicaci\u00f3n. El proceso de inicio es el principal y primer control de la duplicaci\u00f3n del genoma. Trabajos recientes en saccharomyces cerevisiae apoyan que en el inicio de la replicaci\u00f3n del genoma confluyen dos pasos diferentes y consecutivos: deben de formarse complejos pre-replicativos multiproteicos en los or\u00edgenes de replicaci\u00f3n para, despu\u00e9s, ser activados por fosforilaci\u00f3n dependiente de s-cdk y cdc7. La prote\u00edna cdc6 de saccharomyces cerevisiae es necesaria para la formaci\u00f3n de los complejos pre-replicativos que se requieren para la replicaci\u00f3n del dna. Hemos mostrado que la sobreexpresi\u00f3n de cdc6 en cepas deficientes en cdc28 o de una prote\u00edna mutante que carece de sitios putativos de fosforilaci\u00f3n por cdc28 producen re-replicaci\u00f3n del genoma en ausencia de divisi\u00f3n celular. Cdc6 se acumula en algunos mutantes cdc28ts, produce diploidizaci\u00f3n de uno de ellos (cdc28-4) y se estabiliza en cepas deficientes en la ruta de proteolisis dependiente de ubiquitinaci\u00f3n, cdc4-1 y cdc34-2, mostrando que cdc6 se degrada de una forma dependiente de cdc4p\/cdc34p, tras ubiquitinaci\u00f3n, en fase s. Proponemos que la degradaci\u00f3n de cdc6 es parte del mecanismo por el que las c\u00e9lulas de s. Cerevisiae limitan la replicaci\u00f3n a fase s, aunque deben existir mecanismos alternativos mediados por complejos quinasa cdc28-clb previniendo la re-replicaci\u00f3n. Cdc14 es una fosfatasa, con papel en el inicio de la replicaci\u00f3n in vivo ya que limitando su actividad desciende el n\u00famero de or\u00edgenes activos.  nuestros resultados apoyan la idea de cdc14 es parte del control que determina este n\u00famero. Mostramos que cdc14 interacciona gen\u00e9ticamente con cdc6 sugiriendo que ambos genes cooperan controlando la activaci\u00f3n de los or\u00edgenes cromos\u00f3micos en s.
