{"id":35706,"date":"1998-01-01T00:00:00","date_gmt":"1998-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-molecular-de-resistencia-a-beta-lactamicos\/"},"modified":"1998-01-01T00:00:00","modified_gmt":"1998-01-01T00:00:00","slug":"estudio-molecular-de-resistencia-a-beta-lactamicos","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/estudio-molecular-de-resistencia-a-beta-lactamicos\/","title":{"rendered":"Estudio molecular de resistencia a beta-lactamicos."},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Molina Moreno Jos\u00e9 Miguel <\/strong><\/h2>\n<p>En este trabajo se ha estudiado la proporci\u00f3n de las batalactamasas plasm\u00eddicas tem-1, tem-2, y rob-1 presentes en 177 cepas de haemophilus influenzae recogidas en el servicio de microbiolog\u00eda del hospital la fe de Valencia.  basamos el estudio en t\u00e9cnicas moleculares (pcr, frlps, y secuenciaci\u00f3n de \u00e1cidos nucleicos) comparando los resultados obtenidos con los de las t\u00e9cnicas microbiol\u00f3gicas cl\u00e1sicas (prueba del nitrocefin, y estudio de cmi para los antibi\u00f3ticos ampicilina, coamoxiclav, cefaclor y loracarbe). Durante el estudio apareci\u00f3 un gen mutante derivado de blatem-1un 37,2% portaba alguno de los siguientes genes de resistencia :  blatem-1(41,5%), blatem-1(mutante), (46,2%) y blatem-2(12,3%).Un 24,6% de estas cepas portadoras de gen blatem dieron negativo a la prueba del nitrocefin, correspondiento el 62,5% a (tem-1\/tem-2) y el 37,5% al mutado. Esto indica que las cepas con la mutaci\u00f3n son m\u00e1s expresivas, y por tanto m\u00e1s resistentes. Para confirmar esta hip\u00f3tesis se estudiaron estad\u00edsticas las cmls de todas las cepas resistentes genot\u00edpica y\/o fenotipicamente. Al comparar el mutante con tem-1 se observa no hay diferencias significativas para la ampicilina ni para el coamoxiclav, pero si para cefaclor y loracarbef, siendo las cmis del mutado superior que las de tem-1. Cuando comparamos el mutante con tem-2 no hay diferencias significativas. Con respecto a la otra beta-lactamasa ogjeto de estudio, rob-1 hicimos un estudio paralelo. Rob-1 aparecia en el 3,9% del total de las cepas y en el 9,7% de los positivos. Al comparar con los otros grupos resistentes observamos que hay diferencias significativas para todos los antibi\u00f3ticos excepto para el coamoxiclav siendo las cmis muy superiores para las cepas rob-1 un 8,8% de cepas fue positivo al nitrocefin no presenta ninguno de los genes de resistencia estudiados (p.C.R.
